还原变性的溶菌酶的体积排阻色谱复性

Renaturation of Reduced/denatured Lysozyme by Size Exclusion Chromatography

  • 摘要: 探讨二次体积排阻色谱在非氧化还原平衡体系及中性条件下对还原变性的溶菌酶(Lys)的复性.结果表明:在初次排阻色谱的溶菌酶洗脱液中加入过量的氧化型谷胱苷肽(GSSG)和盐酸胍(GuHCl)充分作用后,进行二次体积排阻色谱能够成功复性还原变性的溶菌酶.

     

    Abstract: Renaturation of reduced/denatured Lysozyme in non-oxidation and reduction condition was studied with size exclusion chromatography used twice.The result shows that reduced/denatured Lysozyme can be renatured if excessive oxidized form of Glutathione and Guanidine Hydrochloride are added to elution solution of the first size exclusion chromatography to react completely,which would then be for a second size exclusion chromatography.
