LncRNA SIL Silencing Promoted Proliferation and Migration of A549 Cells

  • 摘要: 研究了lncRNA SIL与肺纤维化中肺泡上皮细胞的增殖和迁移之间的关系.设计小干扰RNA沉默内源性lncRNA SIL的表达,通过RT-PCR、MTT、Western blot、细胞损伤修复实验及Transwell实验研究沉默后细胞的增殖和迁移变化.MTT结果显示:沉默lncRNA SIL后细胞的增殖能力与对照组相比显著增加,细胞增殖相关蛋白PCNA和Ki67的表达量也显著增加.细胞损伤修复实验和Transwell实验结果都显示,沉默lncRNA SIL后细胞的迁移能力与对照组相比显著增加.上述结果说明lncRNA SIL沉默后能够促进肺泡上皮细胞A549的增殖和迁移,lncRNA SIL很可能是潜在的肺纤维化的抑制因子.

     

    Abstract: The relationship between lncRNA SIL and alveolar epithelial cells on the proliferation and migration in pulmonary fibrosis was investigated. The small interference RNA were designed to silence the expression of endogenous lncRNA SIL, the proliferation and migration of cells after lncRNA SIL silencing were studied by RT-PCR, MTT, Western blot, Cell damage repair experiments and Transwell. Compared with negetive control group, the proliferation ability of cells was increased significantly after lncRNA SIL silencing, and the expression levels of protein PCNA and Ki67 were increased significantly, too. The migration ability A549 cells after lncRNA SIL silencing was increased significantly compared with negetivecontrol group by cell damage repair experiments and Transwell. These results showed that lncRNA SIL silencing can promote the proliferation and migration of A549 cells and it is speculated that lncRNA SIL might be a potential inhibitor of pulmonary fibrosis.
