胡颓子叶中槲皮素和山柰酚薄层鉴别及其总黄酮、免疫活性研究

TLC Indentification of Quercetin and Kaempferol and Determination of Total Flavonoids,Invitro Immunostimulatory Activities Analysis of Elaeagnus pungens

  • 摘要: 采用薄层色谱法对胡颓子叶中槲皮素和山柰酚进行定性鉴别,采用紫外-可见分光光度法测定总黄酮,并通过小鼠巨噬细胞(RAW264.7)模型评价其体外免疫活性.结果表明:在薄层色谱中能检测到槲皮素和山柰酚;总黄酮在0.008~0.054 g/L范围内具有良好的线性关系(R=0.9991),回收率为99.6%,RSD为1.32%;胡颓子叶总黄酮能有效促进免疫细胞增殖,可显著抑制LPS诱导的巨噬细胞RAW 264.7中TNF-α的分泌量,表明其具有较好的免疫活性.

     

    Abstract: Quercetin and kaempferol were identified by TLC. The content of total flavonoids was detected by UV. The invitro immunostimulatory activities of Elaeagnus pungens were investigated by using the immune activity analysis cell model (RAW264.7 macrophages).The results showed that:quercetin and kaempferol could be detected by TLC; the calibration curve showed good linearity over the range of 0.008~0.054 g/L(R=0.9991);the average recovery was 99.6% with RSD of 1.32%;the research of immunological activity of the general flavone of Elaeagnus pungens showed that it could effectively promote the proliferation of immune cells, and significantly inhibit the secretion of TNF-α in LPS-induced macrophages RAW 264.7,indicating its remarkable immunological activity.
