血红蛋白作催化剂酸性铬蓝K褪色法测定过氧化氢

  • 摘要: 研究了牛血红蛋白(Hemoglobin,Hb)作为过氧化物模拟酶,以酸性铬蓝K(Acid Chrome Blue K,ACBK)作为氢供体底物的酶催化反应体系的催化特性和反应条件,体系在pH9.6的条件下波长为536 nm处有最大吸收,建立了褪色程度ΔA与H2O2浓度的线性关系,从而建立了测定H2O2的新方法.测定过氧化氢的线性范围为4.0×10-7~3.2×10-5mol/L,检出限为2.7×10-8mol/L.该方法用于雨水和消毒水中过氧化氢含量的测定,结果满意,加标回收率在95.0%~100.0%之间.

     

    Abstract: The catalytic activity of hemoglobin to oxidation of Acid Chrome Blue K(ACBK) by peroxide hydrogen was studied.The catalytic characteristic and reaction condition of Hb-ACBK-H2O2 system were discussed.At pH 9.6,the system exhibited maximum absorption wavelength at 537 nm.The linear range of 4.0×10-7~3.2×10-5 mol/L H2O2 and the detection limit of 2.7×10-8mol/L were obtained.The proposed method was used in the analysis of rainwater and disinfector with satisfactory recoveries of 95.0%~100.0%.
