Synthesis of Adapalene

  • 摘要: 以对溴苯酚和金刚烷醇为原料,在硫酸和冰醋酸催化下,制得2-(1-金刚烷基)-4-溴苯酚(4);化合物(4)用硫酸二甲酯甲基化得2-(1-金刚烷基)-4-溴苯甲醚(5);化合物(5)的格氏化物和6-溴-2-萘甲酸甲酯作用,制得6-3-(1-金刚烷基)-4-甲氧基苯基-2-萘甲酸甲酯(7);化合物(7)在氢氧化锂作用下皂化水解,得到阿达帕林(1).4步反应总收率为71.1%.

     

    Abstract: A Practical synthesis of Adapalene was described.Adapalene was prepared from 4-bromophenol and 1-adamantanol by four steps in 71.1% overall yield.4-Bromophenol was alkylated by 1-adamantanol to form 2-(1-adamantyl)-4-bromophenol(4).The compound 4 then reacted with dimethyl sulfate to give(5).Compound 5 was condensed to 6-bromo-2-naphthoic acid methyl ester(6) to produce 6-(3-(1-adamantyl)-4-methoxyphenyl)-2-naphthoate(7).Saponification of compound 7 methanol and LiOH afforded Adapalene(1).
