艾叶酶协同超声提取工艺优化及其抗氧化、抑菌活性研究

Optimization of enzyme assisted ultrasonic extraction process for Artemisia argyi and study on its antioxidant and antibacterial activities

  • 摘要: 基于层次分析-熵权法综合评分体系,结合响应面试验设计,优化艾叶酶协同超声提取工艺;采用DPPH自由基、OH自由基清除能力测定法测定艾叶提取液的抗氧化能力;利用抑菌圈法和二倍稀释法测定艾叶提取液的抑菌能力。结果表明,艾叶酶协同超声提取工艺最佳参数为纤维素酶用量3.5%、酶解温度55 ℃、酶解时间75 min、超声时间40 min,在此条件下,综合评分为96.65±1.44。艾叶提取液对DPPH自由基和OH自由基的IC50值分别为(45.00±1.43) mg/mL、(47.51±1.14) mg/mL,具有较好的抗氧化活性。艾叶提取液对金黄色葡萄球菌、大肠杆菌和枯草芽孢杆菌的抑菌圈直径分别为(15.20±0.40) mm、(14.10±0.20) mm、(13.16±0.18) mm,最低抑菌浓度分别为0.25、0.50和0.50 g/mL,具有良好的抑菌活性。

     

    Abstract: Based on the analytic hierarchy process entropy weight method (AHP-EWM) comprehensive scoring system, combined with response surface test design, the enzyme-assisted ultrasound extraction process of Artemisia argyi was optimized. The antioxidant capacity of Artemisia argyi extract was determined using the determination method of DPPH and OH radical scavenging ability. The inhibition zone method and the double dilution method were used to determine the antibacterial ability of Artemisia argyi extract. The results showed that the optimal process parameters for the enzyme assisted ultrasound extraction of Artemisia argyi were cellulase dosage of 3.5%, enzymatic hydrolysis temperature of 55 ℃, enzymatic hydrolysis time of 75 min, and ultrasonic time of 40 min. Under these conditions, the comprehensive score was 96.65±1.44. The IC50 values of Artemisia argyi extract for DPPH and OH radicals were (45.00±1.43) mg/mL、(47.51±1.14) mg/mL, respectively, indicating good antioxidant activity. The diameters of the inhibition zones of Artemisia argyi extract against Staphylococcus aureus, Escherichia coli, and Bacillus subtilis were (15.20±0.40) mm、(14.10±0.20) mm、(13.16±0.18) mm, respectively. The minimum inhibitory concentrations were 0.25, 0.50 and 0.50 g/mL, respectively, indicating good antibacterial activity.
