调控猪骨骼肌卫星细胞分化关键lncRNAs鉴定及功能研究

Identification and functional study of key lncRNAs regulating the differentiation of porcine skeletal muscle satellite cells

  • 摘要: 猪骨骼肌发育状况直接影响畜牧业经济收益,针对猪骨骼肌发育问题,探究关键lncRNAs对猪骨骼肌卫星细胞成肌分化的调控作用。采用RNA-Seq方法构建了猪骨骼肌卫星细胞的转录图谱。结果表明:3849个基因在不同分化时期骨骼肌细胞中差异表达,其中3557个为mRNAs,292个为lncRNAs;上述差异基因显著富集在轴突引导和致心律失常性右室心肌病等生物学过程及通路中;3个候选lncRNAs(MSTRG.13015、MSTRG.7609MSTRG.18703)可能在猪骨骼肌卫星细胞分化过程中发挥重要调控功能,MSTRG.13015在猪骨骼肌卫星细胞分化60 h时表达量较高;MSTRG.7609在分化0、48 h时表达量相对分化60 h时较高,MSTRG.18703的表达量高于前两个lncRNAs,且随分化时间增长表达量持续升高。

     

    Abstract: The development of porcine skeletal muscle can directly affect the economic benefits of the livestock industry. Aiming at the problem of porcine skeletal muscle development, the regulatory role of key lncRNAs was explored in the myogenic differentiation of porcine skeletal muscle satellite cells. The RNA-Seq method was used to construct the transcriptional atlas of porcine skeletal muscle satellite cells. The results showed that: 3849 genes were differentially expressed in skeletal muscle cells at different differentiation stages, among which 3557 were mRNAs and 292 were lncRNAs; These differentially expressed genes were significantly enriched in biological processes and pathways such as axon guidance and arrhythmogenic right ventricular cardiomyopathy; Three candidate lncRNAs (MSTRG.13015MSTRG.7609MSTRG.18703) might play important regulatory functions during the differentiation of porcine skeletal muscle satellite cells. MSTRG.13015 had a relatively high expression level at 60 hours of porcine skeletal muscle satellite cell differentiation. MSTRG.7609 had relatively higher expression levels at 0 hours and 48 hours of differentiation compared with that at 60 hours of differentiation. The expression level of MSTRG.18703 was relatively higher than that of the previous two lncRNAs and continued to increase with the increase of differentiation time.
