SSR标记引物及其在识别茶树“信阳10号”上的应用

SSR marker primer and its application on identification of tea plant “Xinyang 10”

  • 摘要: 针对基于表型或生化成分在茶树种质间不易识别“信阳10号”品种问题,采用基于茶树品种“舒茶早”基因组数据,开发SSR分子标记引物对茶树种质基因分型的方法,研究了在茶树种质间识别“信阳10号”。用随机挑选的SSR标记引物,对随机挑选的8份茶树种质基因分型,筛选出多态性SSR分子标记引物SCZSSR03,该引物对188份茶树种质基因分型后显示,茶树品种“信阳10号”的DNA带型同其他187份茶树种质的DNA带型有明显差异。本研究筛选的SCZSSR03分子标记引物可有效识别“信阳10号”,基于该标记建立的识别“信阳10号”分子标记技术,可为利用“信阳10号”品种进行茶树种质资源研究、遗传改良和品种保护提供种质识别技术。

     

    Abstract: Aiming at the problem that “Xinyang 10” is not easily identified among tea germplasm based on phenotype or biochemical components, SSR molecular marker primers were developed to genotype tea germplasm based on “Shuchazao (SCZ)” genomic data, and the identification of “Xinyang 10” among tea germplasm was studied. Eight tea germplasm randomly selected from 188 tea germplasm were genotyped with randomly selected SSR markers and the results showed that the SSR marker primer SCZSSR03 was polymorphic and was selected. Using the primer to genotype 188 tea germplasm,the DNA banding pattern of “Xinyang 10” was significantly different from those of 187 other tea germplasm. The SCZSSR03 molecular marker primer screened can effectively identify “Xinyang 10”, and the molecular marker technology for identifying “Xinyang 10” was established based on the SSR marker, which can provide germplasm identification technology for tea germplasm resource research, genetic improvement and protection by utilizing “Xinyang 10” variety.
