基于环介导等温扩增技术建立非洲猪瘟病毒的快速高灵敏度检测方法
Rapid High-sensitivity Detection of African Swine Fever Virus Based on Loop-mediated Isothermal Amplification

摘要: 非洲猪瘟是严重危害中国养猪业的传染病, 为了快速检测非洲猪瘟, 采用环介导等温扩增技术(LAMP), 根据非洲猪瘟病毒p72基因保守序列设计环介导的等温扩增引物, 然后进行引物筛选和反应条件优化。结果显示, 优化后的反应体系在45 min时可检测低至1 copy/μL的非洲猪瘟病毒, 不与其他病毒核酸样品产生交叉反应, 可以裸眼判断检测结果, 具有很好的灵敏性和特异性。Abstract: In order to solve the problem that African swine fever seriously endangers China's pig industry, a loop-mediated isothermal amplification primer(LAMP) was designed according to the conserved sequence of African swine fever virus p72 gene by loop-mediated isothermal amplification technology, and then the primer was screened and the reaction conditions were optimized. The results showed that the optimized reaction system could detect African swine fever virus as low as 1 copy/μL at 45 min, and did not cross-react with other viral nucleic acid samples, the detection results could be judged with the naked eye, with good sensitivity and specificity.
