基于二房室模型的个体化给药方案设计的建模与仿真

Modeling and Simulation of Individualized Drug Delivery Scheme Design Based on Two-compartment Model

  • 摘要: 利用脉冲微分方程,研究了多次静脉注射给药方式下,二房室模型血药浓度的变化规律,血药浓度在稳态情况下将呈现周期性变化。依据用药安全窗口,探讨了给药方案设计的具体流程和算法步骤。进一步地,结合通过实验测得的24位肿瘤患者单次静脉注射氨甲蝶呤后不同时序的血药浓度数据,运用非线性回归分析的方法对24位患者药代动力学特异性参数进行了估计,并运用患者特异性参数值研究了患者个体之间的相似性和差异性。最后,基于不同患者的特异性参数值,给出了患者的个体化给药方案。

     

    Abstract: The impulsive differential equation was used to study the changes of blood drug concentration for the two-atrioventricular model under multiple intravenous administration. The blood drug concentration changed periodically under the stable condition. Based on the drug safety window, the specific flow and algorithm steps of drug administration plan design were discussed.Further, combined with the blood drug concentration data of 24 tumor patients measured by experiment at different time series after a single intravenous injection of methotrexate, the pharmacokinetic specific parameters of 24 patients were estimated by the method of nonlinear regression analysis. The similarities and differences among patients were studied by using the patient-specific parameter values. Finally, based on the specific parameter values of different patients, the individualized drug administration plan was given.
