山药bHLH基因家族鉴定及表达分析

Identification and Expression Ananlysis of bHLH Gene Family in Chinese Yam(Dioscoreae Rhizoma)

  • 摘要: 用生物信息学和分子生物学方法,在全基因组水平对山药bHLH基因家族进行了系统分析。鉴定了67个基因家族成员,克隆并检测干旱、高盐等逆境胁迫下DrbHLH5的基因表达模式。结果显示,山药bHLH基因家族属于亲水蛋白,成员理化性质差异较大,67个bHLH蛋白可分为17组,同组的成员趋于拥有相同的保守基序和基因结构。该家族基因在染色体上分布不均匀,有些以基因簇存在。三维结构分析表明, 该家族成员具有典型的螺旋-环-螺旋结构。山药bHLH5基因可响应干旱和盐胁迫,暗示其具有潜在抗逆功能。

     

    Abstract: The bHLH gene family of Chinese Yam is systematically analyzed at the whole genome level by the bioinformatics and molecular biology methods. Sixty-seven genes are identified, and the gene expression patterns of DrbHLH5 under drought and high salt stress are detected by qRT-PCR. The results show that the bHLH gene family belonged to hydrophilic proteins. The 67 DrbHLHs can be divided into 17 groups, and the members in the same group tend to have the same conserved motif and gene structure. The genes of this family are distributed unevenly on chromosomes, and some form as gene clusters. Three dimensional structure shows that DrbHLHs have typical helix-loop-helix structures. Gene expression analysis shows that DrbHLH5 can respond to drought and salt stresses, suggesting that it has potential stress resistance function.
