乙烯调控水稻根系生长素合成及运输研究

Research of Ethylene Modulated Auxin Biosynthesis and Transport in Rice Root

  • 摘要: 用乙烯的前体物质乙烯利处理水稻根系,观察根系表型、DR5∷GUS转基因材料GUS活性、生长素的合成基因和生长素运输载体基因表达水平的变化.结果表明:低浓度乙烯利促进侧根伸长;高浓度乙烯利抑制初生根和侧根的伸长以及侧根的起始;乙烯利处理表现出促进根毛的伸长;DR5∷GUS活性在根系中随乙烯利浓度升高而增强;生长素合成相关基因OsYUCCA1、OsYUCCA3、OsYUCCA4、OsYUCCA6上调表达;生长素输入载体基因OsAUX1、OsAUX2、OsAUX4、OsAUX5下调表达;生长素输出载体基因OsPIN5a、OsPIN5bOsPIN8、OsPIN9显著下降表达.

     

    Abstract: Root phenotype, DR5∷GUS activity, and expression of auxin biosynthesis and polar auxin transport genes in rice seedlings treated with ethephon were studied. The results demonstrated that lateral root elongation was promoted in seedling treated with low-concentration of ethephon, and lateral root inition and elongation was decreaed in seedling treated with high-concentration of ethephon. Furthermore, primary root length was shorter and root hair was longer in seedling treated with high-concentration of ethephon compared with control. GUS activity was increased in the ethephon treated DR5∷GUS transgenic plants.Furthermore, auxin biosynthesis genes OsYUCCA1, OsYUCCA3, OsYUCCA4, OsYUCCA6 were up-regualtion, and auxin transport genes OsAUX1, OsAUX2, OsAUX4, OsAUX5, OsPIN5a, OsPIN5b, OsPIN2, OsPIN8, PIN9 were down-regulated in ethephon treated rice root.
