变异链球菌LuxS基因突变株对葡萄糖摄取的研究

A Study on Glucose Uptake by the Streptococcus mutans LuxS Mutant

  • 摘要: 使用预酸化然后培养的方法在一组pH值梯度酸性环境中培养变异链球菌标准株和LuxS基因突变株,比较两种菌株的葡萄糖摄取能力.在pH5.5的BHI液体培养基中预酸化,设定初始pH值6.0~7.0,然后用葡萄糖氧化酶法测定在预酸化和未酸化条件下LuxS基因突变株培养上清液中葡萄糖减少的量.在不同培养条件下,LuxS基因突变株的糖摄取量明显低于亲代菌株(P<0.01);在pH5.5的培养基中预酸化后,突变株的糖摄取量增加(P<0.05).表明LuxS基因突变株的葡萄糖摄取能力降低,其致龋力低于标准株.

     

    Abstract: After preacidification, Streptococcus mutans international standard strain and LuxS mutant strain were cultivated in BHI liquid culture medium with a pH gradient acidic environment in order to study the growth and glucose uptake of Streptococcus mutans after LuxS mutation in vitro. The two strains were preacidificated in BHI liquid culture medium with pH5.5, the initial pH value was set at 6.0~7.0, and the glucose uptake of the Streptococcus mutans LuxS mutant and international standard strain from the BHI medium was measured by the glucose oxidase method under the pre-acidification and non-acidification conditions. The glucose uptake of LuxS mutant strain was significantly less than that of international standard strain at pH6.0~7.0 (P<0.01). The glucose uptake of the LuxS mutant strain from preacidified culture was more than that which did not to be deal with (P<0.05). The glucose uptake ability of the Streptococcus mutans LuxS mutant decreased, LuxS mutant had a weaker cariogenic potential than international standard strain.
