白菜hau CMS不育系特异分子标记的开发与应用

Development and Application of Specific Molecular Markers for the hau CMS in Chinese Cabbage

  • 摘要: 以不同胞质的白菜和芥菜为材料,将hau CMS胞质的线粒体基因组与芸薹属植物中其他的细胞质雄性不育系的线粒体基因组进行比较,再经生物信息学分析,筛选得到hau CMS线粒体基因组上特异的序列,并根据该序列信息开发出用于鉴定白菜hau CMS类型的分子标记hau CMS-Specific.同时,通过序列对比分析发现hau CMS线粒体基因组缺失但其他胞质共有的线粒体基因组序列.根据该序列信息,开发了互补检测的分子标记hau CMS-Deletion.最后利用hau CMS、ogu CMS、pol CMS、oxa CMS、orf220 CMS等不同胞质类型材料的DNA成功验证了这两对标记.

     

    Abstract: Chinese cabbage and leaf mustard were used as basic materials.The mitochondrial genome of hau CMS cytoplasm was compared with the mitochondrial genome of the other cytoplasmic male sterile line used in Brassica, after bioinformatics analysis, the DNA sequence specific to the hau CMS mitochondrial genomes were selected. Based on the sequence information, a hau CMS-specific molecular marker was developed to identify the hau CMS type cabbage. Meanwhile, the hau CMS deleted mitochondrial genome sequence but shared by other cytoplasms was found. Based on this sequence information, a hau CMS-Deletion molecular marker was developed. And the two pairs of markers were successfully verified by using DNA from different cytoplasmic materials such as hau CMS, ogu CMS, pol CMS, oxa CMS, orf220 CMS.
