疲劳模型大鼠T1/T2平衡变化及其与IL-12、IL-27的关系研究

Study on T1/T2 Balance and its Relationship with IL-12 and IL-27 in Fatigue Model Rats

  • 摘要: 选取20只8周龄清洁级SD大鼠,随机分成正常对照组(A组,n=10)与疲劳模型组(B组,n=10).B组采用力竭游泳运动训练的方式建立疲劳模型,在完成最后一次力竭游泳后24 h采集股动脉血,采用ABC-ELISA法分析血清IFN-γ、IL-4、IL-12、IL-27含量.结果表明:(1) B组大鼠血清IFN-γ含量与IFN-γ/IL-4比值均显著低于A组(P<0.05),但血清IL-4含量与A组相比无显著性差异(P>0.05);(2) B组大鼠血清IL-12与IL-27含量均低于A组,其中血清IL-12的比较存在显著性差异(P<0.05);(3)大鼠血清IL-12与IFN-γ含量显著相关(P<0.01),但血清IL-27与IFN-γ含量、IL-12与IL-27含量之间均无显著相关关系(P>0.05).疲劳大鼠出现T1/T2失衡,表现为T1型免疫反应抑制,该变化与IL-12分泌的变化有关,但与IL-27分泌的关系并不密切.

     

    Abstract: Twenty sprague-dawley rats aged 8 weeks were randomly divided into 2 groups:1) control (group A, n=10);2) fatigue model (group B, n=10). Fatigue model was established using exercise training of exhausted swimming in group B. 24 h after the last exhausted swimming in group B, femoral artery blood was collected and the concentrations of IFN-γ, IL-4, IL-12, IL-27 in serum were measured using ABC-ELISA. The experimental result indicated:1) Both of the level of serum IFN-γ and IFN-γ/IL-4 in group B were significantly lower than that in group A (P<0.05), but there was no significant difference of serum IL-4 between group B and group A (P<0.05). 2) Serum IL-12 concentration in group B was significantly lower than that in group A (P<0.05). 3) The concentration of serum IL-12 correlated positively with that of serum IFN-γ (P<0.01), but there were no significant correlation between serum IL-27 and IFN-γ, IL-12 and IL-27 (P>0.05) in rats. In conclusions, T1/T2 imbalance in fatigue rats, characterized by T1-type immunosuppression, is related to production of IL-12, but not closely related to production of IL-27.
