稀土钐离子与牛血清白蛋白相互作用研究

摘要: 利用荧光光谱法研究了稀土钐离子(Sm3+)与牛血清白蛋白(BSA)的相互作用,探讨了不同pH值的缓冲溶液和不同钐离子浓度对BSA荧光强度的影响.通过改变钐离子标准溶液浓度范围,扫描Sm3+与牛血清白蛋白相互作用的荧光发射光谱,证实了稀土钐离子(Sm3+)对BSA的荧光猝灭属于静态猝灭.利用荧光猝灭的Stern-Volmer方程和静态猝灭公式线性拟合,求得反应的荧光猝灭过程速率常数Kq=4.12×1012 L·mol-1·s-1,结合常数Ka=9.652×105 L·mol-1,结合数n=1.22.Abstract: The interaction between rare earth samarium ion and bovine serum albumin(BSA) was investigated by fluorescence spectrum. Effects of the concentrations of rare earth samarium ion and the pH of buffer solutionon the fluorescence intensity were discussed. Fluorescence emission spectrum of bovine serum albumin was scanned with the changing of samarium ion concentration. The results showed that it was a static quenching. Using Stern-Volmer equation and double-reciprocal equation, the quenching constant and binding constant were calculated as follows:Kq=4.12×1012 L·mol-1·s-1, Ka=9.652×105 L·mol-1, n=1.22.
