过氧化氢诱导前列腺癌 P C 3 细胞 H S P 6 0表达和 A k t 蛋白磷酸化的研究

摘要: 以不同浓度 H 2 O 2 刺激 P C 3 细胞 2 4h , 应用 MT T 法分析细胞活力变化, 并采用蛋白印迹方法检测 H S P 6 0 蛋白、 A k t 蛋白( 总 A k t 蛋白) 和磷酸化 A k t 蛋白的表达水平 . 结果表明, 1 5.6 2 5 μ m o l / L 的 H 2 O 2 可诱导 P C 3 细胞内 H S P 6 0 表达显著升高(p 0. 0 5 ) , 7. 3 1 2 μ m o l/ L 和 1 5.6 2 5 μ m o l / L 的 H 2 O 2 均会导致磷酸化A k t 蛋白的量显著下降( p 0. 0 1 ) , 但该两种浓度的刺激对细胞内总 A k t 蛋白量并无显著影响( p 0. 0 5 ) .Abstract: PC3 cells were stimulated with H2O2 in different concentrations for 24 hours,the viability of PC3 cells were measured by MTT assay. Western blotting was used to detect HSP60 protein levels, p-Akt and total Akt protein levels as well.The results of the study confirmed that higher concentrations of H2O2 (15.625 μmol/L) could significantly induce up-regulation of the HSP60 expression ( p <0.05), it also results down-regulation of the p-Akt protein expressing at both 7.312 μmol/L and 15.625 μmol/L concentration of H2O2 ( p <001), but the stimulation of the two concentrations of the H2O2 could not change the total amount of Akt protein levels ( p >0.05).
