柑橘皮中黄酮类化合物与牛血清白蛋白的相互作用

Interaction between Bovine Serum Albumin and Flavonoids from Orange Peel

  • 摘要: 用甲醇作为溶剂,提取柑橘皮中黄酮类化合物.在模拟人体的生理条件下,采用荧光光谱法研究牛血清白蛋白(BSA)与黄酮类化合物的相互作用.结果表明,黄酮类化合物浓度在1.0~10.0μmol.L-1范围内,它与牛血清白蛋白的荧光强度之间符合关系式If=-34.552c*+434.66,相关系数R2=0.9308.两者之间有一个结合位点,结合常数为5.18×105L.mol-1,猝灭常数Kq=3.99×1013L.mol-1,相互作用过程属静态猝灭.

     

    Abstract: Flavonoids were extracted from the orange peel using methanol as solvent. The interaction between bovine serum albumin (BSA) and flavonoids was studied by fluorescence spectroscopy under the physiological conditions of simulated human body. The linear ranges were 1.0~10.0 μmol·L-1and the linear equationI f =-34.552c*+434.66 and R2 = 0.9308. The binding constant for the binding site between BSA and flavonoids was calculated to be 5.18 × 105  L·mol-1. The quenching constant was calculated to be Kq = 3.99 × 1013L·mol-1.
