不同培养基和生长调节剂对细叶小羽藓生长发育及愈伤组织诱导的影响

Effects of Different Culture Medium and Growth Moderating Substances on Growth and Callus Induction of Haplocladium Microphyllum

  • 摘要: 对淮河上游南湾湖周边的细叶小羽藓(Haplocladium microphyllum)的孢子生长发育进行观察,发现细叶小羽藓孢子无休眠现象,孢子接种2天左右萌发;扩大培养实验结果表明,细叶小羽藓无菌原丝体在1/2 MS培养基上置于20℃±2℃、12 h光照/12 h黑暗条件培养,产生配子体最多,且配子体长势最好,可以获得大量无菌材料;细叶小羽藓愈伤组织诱导实验显示,形成愈伤组织的最佳培养基为添加1%葡萄糖+1.5 mg/L 6-BA+0.25 mg/L 2,4-D的MS培养基.

     

    Abstract: Based on observation of the growth and development of spores of Haplocladium microphyllum in the south of Nanwan lake of Xinyang, it was found that there was no dormancy in the sphagnum spore, and the spore inoculation was germinated about 2 days. Expand training experimental results showed that the fine Haplocladium microphyllum sterile protonema moss on the 1/2 of the MS culture medium at 20℃±2℃, 12 h of light/dark conditions to cultivate, produce new stem leaf body up, and the stem leaf do best, get a lot of aseptic materials; The experimental results showed that the optimal medium for the formation of callus was 1% glucose + 1.5 mg/L 6-BA+0.25 mg/L 2,4-D MS medium.
