α-淀粉酶协同超声波法提取复方半边莲注射液工艺优化

Optimization on Extraction Technology of Compound Lobelia Injection by Synergistic Action of α-amylase and Ultrasonic Method

  • 摘要: 在超声波条件固定的情况下,采用正交试验设计,运用α-淀粉酶提取复方半边莲注射液指标成分野黄芩苷,以HPLC方法检测野黄芩苷含量.最佳工艺条件为:酶解时间30 min,pH6.0,酶解温度30℃,酶用量0.5 g.在此优化条件下,提取复方半边莲注射液的野黄芩苷含量为4.45 g/L,明显大于复方半边莲注射液注册标准煎煮法野黄芩苷的含量1.34 g/L、单一超声法3.84 g/L、单一α-淀粉酶法1.89 g/L.

     

    Abstract: The scutellarin of compound lobelia injection was extracted by the intergrated treatment with α-amylase and ultrasonic,and was determined by HPLC in orthogonal experiment.The optimal conditions were the following: the enzymolysis time is 30 min, the optimal pH is 6.0,the enzymolysis temperature is 30 ℃ with the α-amylase dosage is 0.5 g . Under such optimized conditions, the content of scutellarin in compound lobelia injection is 4.45 g/L, which is significantly more than 1.34 g/L through decocting technology contained in registration standards,3.84 g/L through single ultrasonic extraction, and 1.89 g/L trough single α-amylase extraction.
