The Separation,Purification and Identification of Duck Liver s Metallothionein

  • 摘要: 以信阳华英鸭为材料,用ZnSO4诱导其肝脏合成金属硫蛋白(MT),将鸭肝材料经Sephadex G-50柱层析和DEAE-Sepharose CL-6B阴离子交换柱层析2次分离纯化,得到Zn-MT,该MT由61个氨基酸组成,其中半胱氨酸含量约为28%,分子量6.5 KD.鸭肝Zn-MT产率为0.57 mg/kg鲜肝重,在220 nm有吸收峰.

     

    Abstract: The material of this experiment is Huaying duck from Xinyang area.The synthesis of metallothionein can be induced by ZnSO4 in the duck liver.The Zn-MT was isolated and purified by Sephadex G-50 column chromatography and DEAE-Sepharose CL-6B negative ion exchanging column.The protein is composed of 61 amino acids,including 28% of cysteine.The molecular weight of the protein is 6.5 KD,and the absorbing peak is shown at 220 nm.The yield rate of Zn-MT is 0.57mg/kg of the fresh liver.
